if I have Daylight scheduled to backup at a certain time, and Daylight is not running at that time, will the backup still occur?

Yes; the backup process is handled by the database engine (OpenBase); as long as the database is running, it will be backed up at the scheduled time.

Mine isn't backing up, even though it's set to. I always had that feature enabled in openbase directly, and it worked great. It's broken since 3.7.6.

The new preferences in the Maintenance pane simply reflects/sets the settings in OpenBase; check the settings in OpenBase to see if they match what Daylite displays (remember that the settings displayed in Daylite can only apply to the local database on that same computer).

The DL preference and openbase setting match each other on the server computer. But it doesn't back up. It did a nightly backup until 3.7.6.

I just checked and mine hasn't backed up in several days...
